Littrell, who took over after former offensive coordinator Jeff Lebby left for a head coaching job at Mississippi State, will be dismissed after just seven games.
After being elevated from his offensive analyst position ahead of the 2024 season, OU is currently ranked 93rd in scoring offense and 125th in total offense nationally.
The firing comes after OU's 35-9 loss to South Carolina on Saturday.
"Seth is an all-time great Sooner," OU head coach Brent Venables said in a press release on Sunday. "He has a deep love for this university and football program, and has poured his heart and soul into both. Despite that, our performance as an offense this season has not at all lived up to the OU standard and I felt a change was necessary now."
Littrell, who is a Muskogee native was a fullback on OU's 2000 national championship team, currently has a contract that runs through 2026 and will pay $1.1 million per year.
SoonerScoop also noted that current co-offensive coordinator and tight ends coach Joe Jon Finley, who was in the booth for the first time against South Carolina, will call plays moving forward.
"The leadership role Joe Jon plays on our team is critical," Venables said. "He has the confidence of our locker room and coaching staff, and I'm thankful to him for taking on this expanded role in the middle of the season."
Finley, who was both a player and graduate assistant at OU, joined the Sooners in 2021 as an assistant head coach for offense and a tight ends and half back coach. He was promoted to co-offensive coordinator last November.
Offensive analyst Kevin Johns will join Finley as the quarterbacks coach.
"Kevin joined our staff last spring as an offensive analyst after serving as an offensive coordinator for over a decade," Venables noted. "He has a wealth of experience coordinating offenses and coaching quarterbacks, and I'm looking forward to the fresh approach he'll bring and the bigger role he'll play in offensive game-planning."
The two will step into their new roles ahead of OU's game against No. 18 Mississippi on Saturday.
"I'm confident Joe Jon, Kevin and the rest of the offensive staff will work tirelessly to put our players in positions to succeed and make us more effective on that side of the ball," Venables said.
